How do you use RNase-free DNase set?
Dissolve lyophilized DNase I (1500 Kunitz units) in 550 μl of RNase-free water. To avoid loss, do not open the vial. Inject RNase-free water into the vial using an RNase-free needle and syringe. Mix gently by inverting.
Can I Vortex DNase?
one vial of DNase I. Pipette up and down gently to mix. Do not vortex. If needed, aliquot the DNase I into smaller portions and store the unused portions at -20ºC.
At what temperature does DNase denature?
70 °C
Heat at 70 °C for 10 minutes to denature both the DNase I and the RNA. Note: This product should not be used for digestions longer than 15 minutes or for digestions at temperatures higher than room temperature, or the residual contaminating RNase activity will begin to degrade the RNA.
How quickly does DNase work?
One unit of DNase I, Amplification Grade, completely digests 1 ug of plasmid DNA to oligonucleotides in 10 minutes at 37 °C.
How much DNase should I use?
Use 0.01 to 1 mg/ml DNase I. For each cell type, the working concentration must be determined individually. For optimal enzyme activity, add 5 mM Mg2+. DNase I is prepared from bovine pancreas.
What does DNase treatment do?
DNA-free™ DNase Treatment & Removal Reagents contain RNase-free DNase, and an optimized DNase digestion buffer, to ensure safe, complete removal of contaminating DNA from any RNA sample.
Is DNase active at room temperature?
The DNase Max Kit is stable at room temperature (15–25°C) for up to 6 months or at 2–8°C for 2 years with no loss of activity. Room temperature storage eliminates the need to aliquot and freeze stocks of DNase I enzyme and removes concerns about decreased enzyme activity due to freeze–thaw cycles.
How stable is DNase?
Storage/Stability DNase I retains activity for at least three years when stored at –20 °C. can lose ~20% activity. DNase I remains active in solution between pH 5 and 7 up to 60 °C for at least five hours.
How long is DNase good for?
18 months
Store up to 18 months at −15 to −25°C. The solution will not freeze. 20 mM Tris-HCl, 1 mM MgCl2, 50% (v/v) glycerol, pH 7.5. Store up to 18 months at −15 to −25°C.
Is DNase stable at room temperature?
How long does DNase last at?
type I DNAse activity lasts about 6-12 hours (rather count 6), at 37°C pH 7.4.
